The strongest supporting study is Wing 2015, a PET brain-imaging study of 10 people published in Neuropsychopharmacology. It measured [11C]FLB-457 PET cortical D2/3 binding potential after overnight abstinence vs reinstated smoking in nicotine-dependent daily smokers (cortical dopamine release).

In the authors' words: “The overall peak t was located in the cingulate gyrus, which was part of a larger medial cluster (BPND change -12.1±9.4%) and this survived false discovery rate correction for multiple comparisons.”

Evidence grade: A, human imaging or trial evidence with a measured effect.

How the app explains it

Nicotine hits α4β2 receptors on VTA dopamine neurons, pushing fast bursts of firing into the NAc within seconds of each puff. Chronic smoking is linked to reduced D2/D3 availability on PET imaging and the flat, blunted mood smokers feel between cigarettes. Sensitivity typically climbs back across weeks of abstinence.

Pathway: nAChR→VTA burst, D2/D3 downregulation

This is the mechanism card shown on this habit in the app. It is editorial copy written from the studies below; where it and a study disagree, the study wins.

What this means, and what it doesn't

What it means

  • There is published research on how smoking relates to the brain's reward system, and every study it rests on is listed below with a link.
  • At least one human brain-imaging study or trial measured an effect. That is the strongest kind of evidence in this catalogue.
  • If smoking is costing you sleep, focus or time, cutting back and tracking it is a reasonable experiment. How you feel over a few weeks is the result that matters.

What it doesn't

  • It does not show that a "dopamine detox" resets anything. None of the studies listed here tested a detox programme.
  • It is not a measurement of your brain. The app's score for smoking is an editorial estimate: studies support the mechanism, they do not validate the number.
  • It is not a diagnosis or medical advice. If smoking feels out of control, a doctor or counsellor can help.

  • Main study·Human PET imaging · n = 10

Measuring cigarette smoking-induced cortical dopamine release: A [¹¹C]FLB-457 PET study

    Wing 2015, Neuropsychopharmacology · doi:10.1038/npp.2014.327

Measured: [11C]FLB-457 PET cortical D2/3 binding potential after overnight abstinence vs reinstated smoking in nicotine-dependent daily smokers (cortical dopamine release)

Reported effect: 12.1% decrease in [11C]FLB-457 BPND, medial cingulate cluster (-12.1 ± 9.4%; decrease = dopamine release)

“The overall peak t was located in the cingulate gyrus, which was part of a larger medial cluster (BPND change -12.1±9.4%) and this survived false discovery rate correction for multiple comparisons.”

    Dongelmans 2021, Nat Commun · doi:10.1038/s41467-021-27268-7

Measured: Midbrain dopamine neuron tonic activity and exploration/exploitation strategy under chronic nicotine; optogenetic mimicry

“We then mimicked the effect of chronic nicotine on the tonic activity of dopamine neurons using optogenetics, and found that photo-stimulated mice adopted a behavioral phenotype similar to that of mice exposed to chronic nicotine.”

Measured: [11C]raclopride PET striatal BPND after nicotine gum vs placebo; double-blind randomized placebo-controlled crossover in smokers and non-smokers

“Smokers showed significant decrease in BP in the striatum following nicotine administration, but non-smokers did not show such a decrease.”
